Indications
It's used to treat fungal corneal ulcers and keratitis.
Pharmacology
Fluconazole works as a fungicide. In susceptible fungi, it inhibits cytochrome P-450 14-demethylase, causing an accumulation of lanosterol and a decrease in ergosterol concentration, altering the cellular membrane, resulting in increased membrane permeability, leakage of essential elements, and impaired precursor molecule uptake to DNA.
Dosage & Administration
Instill 1 drop to be instilled into the affected eye(s) 5 times daily.
Interaction
Fluconazole can affect the pharmacokinetics of medicines that are metabolized in the liver.
Contraindications
Patients who are hypersensitive to azoles should avoid taking the medication.
Side Effects
This medication is usually well tolerated. Some patients have been diagnosed with eosinophilia.
Pregnancy & Lactation
Use during pregnancy: Category C pregnancy. In pregnant women, there are no sufficient and well-controlled trials. Only if the possible benefit outweighs the risk to the fetus should fluconazole be used during pregnancy.
Use in lactation: The drug is excreted in breast milk in a concentration similar to plasma, hence it should not be given to a nursing mother.
Precautions & Warnings
Fluconazole may cause non-susceptible candida strains other than Candida albicans to overgrow.
Storage Conditions
Keep out of children's reach. Store away from heat and direct light in a cool, dry location. After opening the bottle, do not use it for more than 4 weeks.
